Recognizes a 33 kDa glycoprotein, identified as Nucleophosmin (NPM). It is predominantly localized in the nucleus of cells in most tissues. NPM is involved in ribosomal assembly and rRNA transport. It is an abundant protein that is highly phosphorylated by Cdc2 kinase during mitosis. This phosphoprotein moves between the nucleus and the cytoplasm. It is thought to be involved in several processes including regulation of the ARF/p53 pathway. A number of genes are fusion partners, in particular the anaplastic lymphoma kinase gene on chromosome 2. Mutations in exon 12 affecting the C-terminus of the protein are associated with an aberrant cytoplasmic location. Mutations in this gene are associated with acute myeloid leukemia. This antibody recognizes a sialoglycoprotein of 27-32 kDa, identified as CD99, or MIC2 gene product, or E2 antigen. MIC2 gene is located in the pseudo-autosomal region of the human X and Y chromosome. MIC2 gene encodes two distinct proteins, which are produced by alternative splicing of the CD99 gene transcript and are identified as bands of 30 and 32 kDa (p30/32). Although its function is not fully understood, CD99 is implicated in various cellular processes including homotypic aggregation of T cells, upregulation of T cell receptor and MHS molecules, apoptosis of immature thymocytes and leukocyte diapedesis. CD99 is expressed on the cell membrane of some lymphocytes, cortical thymocytes, and granulosa cells of the ovary. Most pancreatic islet cells, Sertoli cells of the testis, and some endothelial cells express this antigen. Mature granulocytes express very little or no CD99. Down regulation of PMEPA1 is observed in prostate tumors, suggesting that PMEPA1 may exhibit activity as a tumor suppressor. This antibody recognizes a protein of 42 kDa, which is identified as Alpha-methylacyl-CoA Racemase (AMACR), also known as p504S. It is an enzyme that is involved in bile acid biosynthesis and beta-oxidation of branched-chain fatty acids. AMACR is essential in lipid metabolism. It is expressed in cells of premalignant high-grade prostatic intraepithelial neoplasia (HGPIN) and prostate adenocarcinoma. The majority of the carcinoma cells show a distinct granular cytoplasmic staining reaction. AMACR is present at low or undetectable levels in glandular epithelial cells of normal prostate and benign prostatic hyperplasia. A spotty granular cytoplasmic staining is seen in a few cells of the benign glands. Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI 6.0) subfamilies. This antibody recognizes a protein of 145 kDa, identified as CD11c. CD11c (ITGAX), a member of the leukointegrin family, shares the same beta subunit with other members of the leukocyte adhesion molecule family, which includes CD11a (LFA-1), CD11b (MAC-1) and CD11d (ITGAD), but has a unique alpha chain. CD11c has been shown to play a role in phagocytosis, cell migration, and cytokine production by monocytes/macrophages as well as induction of T-cell proliferation by Langerhans cells. CD11c is expressed prominently on the plasma membranes of monocytes, tissue macrophages, NK cells, and most dendritic cells (DCs). A lower level of expression is also observed on neutrophils as a result of its high level of expression on most DCs. An antibody to CD11c may aid in identification of lesions with histiocytic origin. This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently.
